Bremelanotide was evaluated in two randomised phase 3 trials (RECONNECT) in premenopausal women with hypoactive sexual desire disorder, with a long-term extension and safety analyses. A 2019 review in Drugs reports its first approval in the United States.

What adverse events have studies of PT-141 reported?

In phase 3 trials in premenopausal women with hypoactive sexual desire disorder (1,247 in the safety population), nausea, flushing and headache were more common than with placebo; in the extension, drug-related rates were about 40%, 21% and 12% (PMIDs 31599840, 31599847). Focal hyperpigmentation with repeated daily dosing and small transient blood-pressure rises were reported (PMID 35147466).

How do you reconstitute and store PT-141?

Store the sealed vial frozen (about -20 degrees Celsius), dry and away from light until it is reconstituted.

After reconstitution with a sterile solvent, keep the solution at 2 to 8 degrees Celsius and avoid repeated freezing and thawing. Record the solvent, volume and date on the vial label.
